Abstract

The definition of „atipicality” remains unclear, althoughit is generally accepted that, at a clinical level, diminished EPSand a reduced capacity to elevate plasma prolactin levels arecentral to the definition: broader definition include efficacy intreating negative symptoms and refractory patients,pharmacological properties (neurotransmitter receptor affinityin CNS limbic selectivity and dystonic liability). Clozapinebecame the archetype for a new generation of antipsychoticdrugs, which includes quetiapine, olanzapine, risperidone,sertindole, zyprasidone, zotepine and amisulpride. Similaritiesand differences amongst this new agents conclude that there isnot a unique features of atypicality and require controlledclinical trials in addition to preclinical and animal models toclarify it.
