Abstract

The long struggle to isolate the insulin for therapeutic use centered on the confrontation between two scientific paradigms. First, the physiological approach, invented by Kleiner and Paulescu. In their experiments, these researchers used salted water to extract the active principle from pancreatic tissue, in order to reproduce, as much as possible, the way in which the hormone is made available in the living organism. The paradigm enabled Paulescu to discover the physiological properties of insulin. However, the isolation of insulin for clinical use was accomplished by scientists who adopted the biochemical approach, namely Zuelzer, Banting and Best, and Collip, who used organic solvents for the purification of the pancreatic extract.
