Abstract

The quinolones (QN) are a family of synthetic broad-spectrum antibacterial agents, with a 4-oxo-1,4- dihydroquinolinic structure, based on the model of nalidixic acid. The quinolone structure with the 4-oxo and 3-carboxyl groups offers the possibility of chelation between QN and metallic ions. The present paper presents modern theories regarding the complexation phenomenon of QN with metallic ions focusing on the: characterization of complexes (physicochemical characterization, influence of pH, complex solubility, complex stability, behavior of complexes in different biological environments), bioavailability of QN complexes and improvement of their bioavailability, development of new pharmaceutical formulations in order to increase bioavailability and therapeutic effect, antimicrobial effect of the new QN complexes, enlargement of the therapeutic area of QN complexes taking in consideration the mechanism of action especially regarding their selectivity at the bacterial DNA level and development of new analytical methods used for the determination of QN from complexes.
